cutting a wedge of clean pan spores on agar to a LC

Recommended Posts

A couple years ago I made some prints of pan cyan. Almost a week ago I made up a fresh batch of agar and transferred one of them to several dishes, they may be too dry because I don't see any growth from them nor do I see any contamination. I thought I might be able to cut the wedge of spores out and drop them into a liquid culture to get them rehydrated, I've always had a lot of success with liquid cultures from agar and don't see why it won't work. I just sterilized the liquid cultures with stir bars to keep them agitated. Just wondering if anyone has done the same thing or any thoughts or concerns.

To hydrate your spores, I would just make a spore syringe and put 1 drop on each agar dish after 24hrs of making the syringe. Whenever I see no growth after a couple weeks, it always smells sour or gross when I finally open it up to throw it out and start over. Contamination might be present even if it's not visible.

I have very rarely had luck with transferring spores to agar without hydrating them first, as magicman mentioned. You can drop that wedge into LC, but it is very easy to contaminate LC. It wont hurt to try esp if you have the proper equipment and aseptic technique

Yippie my dishes are showing signs of life though to be on the safe side I made a syringe and inoculated a few dishes with it as well. Also I had a syringe I made of a liquid culture I kept in the fridge that I squirted some onto a dish that looks kinda funky do you think it's contaminated? Thanks for the help I'll keep you updated on the progress.

I just picked like 140gs wet. And I'm going to keep the fan blowing on it all night in an attempt for evaporation, I can only imagine my grow when I do this correctly. 😂😂😂😂😂 Those were going to be large boomers 😢😢😢

It’ll do much much better if you can find a way to incubate that bag at 80-82f. You wanna get that fugger colonized as quickly as possible. Plus the longer it sits , the more things can lose moisture content. Otherwise, I would only give the colonizing grains one gentle fondling mix at about 20% or so then leave it be. A second mix/shake at 70% is unnecessary and  can actually stall things

You gotta pick those fruits immeadiatly then. You’ll be ok, as long as you haven’t used chlorinated/ flouridated tap water (it’s a destroyer in my experiences). If it’s pure water , let that water soak in for a few hours or so, then do your best to carefully dump it out.Maybe lean the tub on end for a bit.  I always recase my subs with  fresh 50/50 Casing Layer, because I’ve washed away my prior casing with my Dunk process.

Thank you Phish!!!!  I may have of fucked it all up by dousing the whole thing with water already 😂😂😂😂😂 I'm letting a pretty high powered fan blow into the tank rn.

Keep an eye on those fruits and pick them when the veils begin to break. Likely within the next day or so. Don’t expect any more pins on your first flush. That’s it.  Mushrooms are 90-92% water weight. A heavy flush will leave a sub depleted of moisture and drier than a popcorn fart. You, my friend, don’t need to be concerned with that with this flush. I would do exactly like MZ says with the spray bottle on that sub. That sub should have a lotta Wooooo left to go 😉👍
