  1. So without nutrient agar for the mycelium to feed on does it then turn upon itself to solidify and consolidate its advances?This is my first foray into agar and Petri and just really trying to get a grasp on it all. https://imgur.com/gallery/T3lB5kT
  2. Could someone tell me if this is mycellium growing ? It looks to have dark rings in it, im guessing it is contam or how else would I have gotten such a clen strain from print (used sterile innoculation loop to gather spores from print) PE #6 on Potato Dextrose Agar
  3. This is my first agar Clone from a nice PE cluster. Transfered this over a month ago . The blue piece is the tissue. Is it supposed to look like that ?
  4. So a friend of mine gave me a few syringes that he had lying around and *spoiler alert* they were not stored properly . They were just left with their needles on instead of the leur lock . They’re less than a year old so I figured I’d try and make something happen . The chitwan Ss had just a little juice left in it so I mixed up some agar and then transferred that to a half dozen other jars and then let it cool overnight to help with the condensation . The condensation turned out to be very light on my second attempt . ok so a week ago I put the chitwan on agar and the results were less than stellar . Can’t say for sure that any of this is even myco. As always your comments, put downs and critiques appreciated . I took a couple small pieces and transferred them to new agar jars . Guess we will see if it’s smallpox or mycelium in a few days .

    Didn't understand the splotchy growth, possibly from injecting the agar vs a drop on top. It is hard to see in the airbag.

    Fresh Spores on the loose

    Spores taken open air, looking better than I thought 💭 . 48 hour print and then scraped a bit onto agar. Got DNA 🧬 for years. Making new prints in a sterile jar to make syringes and kickstart this shit. Also have clone of a big mush out a cluster of 13 colonizing. Will post that when it’s come to fruition. spores on day 3 they look like Hawaii 😂 👀

Have a clone from a mushroom growing on these plates and I’m wondering if this growth is looking normal so far mainly how the transferred piece is fluffy and then it seems to get thin as f (3D->2D?) this is day 4/5 and if it’s alright to use micropore tape vs Saran/ cling wrap instead of Parafilm for now Any thoughts?

    Is it the spores or the agar?

    So I'm very new to agar and wanted to share this with the group in Hope's that I can get some info from the more experienced cultivators. The other day I made my first sleeve of agar dishes I used 15g LME and 10g of agar and 500ml of tap water. I pressure cooked for 40min and allowed to cool. I poured the dishes in a SAB and then individually wrapped them in plastic wrap. The next day I decided to inoculate them with some spores I had from previous grows. 3 days later I started seeing signs of life. Unfortunately the wrong kind of life. It looks nothing like the 2 types of mycelium we all go for🤣 I'm not sure if it's bad spores or agar? The agar in the dish with the green spots looks healthy, the growth obviously doesnt. After looking through my other agar dishes to try again, I found a disturbing site. A fully green dish. The dish wasn't inoculated with anything!! I repeat the great monster wasn't inoculated with anything! It just turned this way after 3 days of storage. My dishes have been stored in a consistent 75 degrees Fahrenheit. Here are my questions. 1. Are the spores bad based off what you see in dish one? Should I chuck the whole print I inoculated it with? 2. Is the agar bad? Why would one just turn into a green mess after 3 days? 3 is there to much moisture in my dishes? I let the agar cool so much that it started to clump when I was pouring the dishes. 4. If I let it cool that much and I still got that much condensation how do I prevent this with future dishes? Below are the pics of the condensation, the green monster and the contaminated dish. Thanks for any and all info. I've done lots of grain jars with multi-spore syringes and never had contamination and though thoes grows were good I'm fully committed to agar isolation to make the future generations thicker flushes but for now it looks like I got lots to learn about agar. Any info that can spare me more waste would be amazing. Thanks guys! Take your spirit veggies!
  12. Can some veterans share their easiest agar recipes for a first timer. I had a recipe using telephone brand agar but lost it. I have already bought the agar and now I don't know what the hell to do with it. Im a hands on learner, so links for videos are awesome,,, I need how to step by step.... thanks y'ALL!

Hey folks, cloned the insides of a fresh fruit from a harvest I didn’t expect to fruit but did, cloned it 10 days ago and I had originally wrapped it in glad cling wrap, I think it wa struggling to breathe so I now put micropore tape and am wondering if I should just make a couple transfers from this, should I inoculate a jar then g2g, or just make a new clone or wait on this and see if it takes off, looks fin e, also the piece wasn’t straight and it was a little wet when I cloned. Thanks guys
  14. So a while back SWIM had some rye berry jars that stalled. SWIM never tossed them because they had some nice looking tomentose mycelium in them. So SWIM figured SWIM would mess with some antibiotic Agar. SWIM just poured some plates the best SWIM could and dabbed some that fluffy goodness on them. 6 days later and this one looks like it’s trying to pin(?) SWIM will just isolate the good ryzomorphic growth (you can see it flaring out from the dabs) 3 or 4 times and then inoculate some grain. The antibiotic agar should help cut down/isolate the baddies. ~ Adult Swim

  16. So, this is my 3rd transfer of Puerto Rican agar. Each plate ( do 2 new plates from each ), . I'm thinking this myc has to be tomentose based, or confusing bacteria. Every plate looks exactly the same, never gets rhizomes. Or is it bacterial? Perfect uniformity, doesn't get thicker ( but thick to begin with), super white). Now, on one plate, there seems to be a pin sized drop of possible metabolites, very light, transparent yellow, sitting on top of the myc( too small to photograph). Anyone grown Puerto Rican? Thoughts if this , myc or bacteria? Tired of doing transfers, no blatent sectors, a perfect disc shape. It's thick in the middle, transfered agar was a bit thick.
  17. Hey guys and gals! starting my first grow soon, doing a WBS tek. Got my spawn jars all ready and waiting. What I was wondering, i've tried to make spore prints from store bought portabellos, just to get some practice, and its been 2 days and I cant see a print. on one I think I can see a few faint markings, but its mostly just condensation (but I have heard that sometimes store bought can be tough to grow because of selective breeding etc). and instead of wasting possible spores, or even a spawn jar contam, i wanted to start it on some agar, and just see what would happen. the stores near me are suprisingly out of agar??? but they have plain gelatin, could plain gelatin work? would i have to add sugar or anything like nutrients to supplement the gelatin? are the portabellos going to drop spores? when I bought them the veil was closed, i manually cut the stem and on some, cut parts of the veil off to open it up. thank u everyone! EDIT: going to try a LC by running purified water over the condensation on the tin foil into a pre made LC
  18. hello im new to agar and have isolated what i believed to be mycelium, however its not fuzzy and the sectors look different than what ive seen online, this is a few days after transferring. started with multi spore lc to grain then plicked out some grain from the best looking jars. is it supposed to be slimy or am i growing bacteria

    Was moving out some old agar dishes and had them by the sink for like a week, to clean out and when I finally got to cleaning.I seen this sample alive and strong. Then put to agar and tossed in jar. We will see how it goes.

  21. I'm a newbie and I'm extremely interested in doing my first grow. I have some amazing mushrooms and they are actually dried up already. I would actually like to save the strain or whatever species it is that I have. Is it possible to extract viable spores from a dried closed cap with a syringe by injecting distilled sterile water into the cap and sucking it back up into the syringe? Technically the Cap hasn't opened up and is closed and has not been exposed to the air, although its crispy and dry. I heard of someone who succeeded with a dried closed mushroom cap of a shitake mushroom, and I'm not sure this will work. I'm also deciding on sprinkling some of the dried gills into agar and begin isolating from there, but there are so many different agar recipes and I'm not sure whether I would succeed with a general MEA agar recipe or use an antibacterial agar mix instead since they are dried mushrooms. Any suggestions would be greatly helpful.
  22. Here are 2 different agar plates. I used a single piece of colonized rye, from MS Syringe, rye from same jar. Seems as though I'm seeing 2 types of myc ( without considering the possible contam) first photo shows more of a " pitch-fork type myc ( each strand is branching out), and the other plate, just straight forward single strands. The additional growth spots, could be myc, as the rye rolled around on the agar before I settled it in the center. At this point I'm just curious about the 2 different myc. These were practice dishes. Gonna be doing tissue culture for clones. Probably just gonna toss when fully colonized on the plate. Overall, does the myc look healthy? Based on the photos, would you pick one over the other for future spawn runs? Not too sure what to be looking at / for regarding quality myc. Any opinions would be appreciated. Again, just practicing, and not concerned about the possible contam.
  23. New to growing here. I want to start getting in to agar work but don't have my petri dishes....or agar...or flow hood setup. One of my golden teacher multispore grows is huge and i want to clone the sucker and it has to be tomorrow. However, since i dont have the agar I was wondering if i could clone to LC first. Let it grow and do its thing, then grab a sample and put it back on agar to check for, and separate from possible contams. I"m thinking this way i can store my LC in the fridge until I have my flowhood set up and agar in hands. So the idea here is to grab a clone of this fruit (whos veil will probably break tomorrow) and preserve it in LC until I have my agar ready to check it for contams and actually grow. So far my reading brings me to beleive that the best sort of long-term storage of a clone sample is with LC in the fridge. I have heard you can keep it in the fridge for 6 months+. Can you keep a petri dish with agar in the fridge for that long without the myc becoming unusable?

    So I've done some agar work and noticed a yellow spot next to the myc. Is this bacteria or what is it ? And should I just transfer the myc to another glass ?:) I took some pictures of some other agar jars, with some spots I noticed aswell. What is this ? - happy growing!
  25. Hello all, another nub with a question I’m sure have been asked and answered. I’ve searched for and could not find a comprehensive Supplies list to perform isolation Tek on a grow. It would be greatly appreciated if someone could please point me in the right direct. I’m looking for details of what type of tubes work best. What kind of media do you pc agar in. What size syringe and needle and where do you buy them. Thank you all.
